| 内容記述 | 本研究では、ポリグルタミン（PolyQ）病、筋萎縮性側索硬化症（ALS）などの神経変性疾患における原因蛋白質の液-液相分離（LLPS）の制御破綻による凝集メカニズムの解明を目的として、１）Thio-PolyQとTDP-43-MBPの試験管内実験系を樹立し、両蛋白質のLLPS・凝集を阻害する化合物Xを同定した。２）培養細胞において、DCTN1ノックダウンなどの微小管輸送障害によりTDP-43のストレス顆粒からの解離が遅延し、さらにALSモデルショウジョウバエにおいて、DCTN1など関連分子のノックダウンによりTDP-43凝集体および神経変性が増悪することを明らかにした。 | |||||||||

We identified compound X that can inhibit LLPS and aggregation of both Thio-PolyQ and TDP-43-MBP. We also found that disruption of microtubule-dependent transport by DCTN1 deficiency delays the disassembly of TDP-43 stress granules in cells, and accelerates TDP-43 aggregation and neurodegeneration in ALS Drosophila models. | |||||||||
