内容記述 | [Synopsis] The increase of sphinganine/sphingosine (SA/SO) ratio in biological fluids and tissues is currently adopted as biomarker to display early toxicity and exposure to fumonisins, mycotoxins largely occurring in maize worldwide and produced by Fusarium moniliforme and F. proliferatum. Results of the recent work carried out at our Institute with this biomarker is reviewed herein. We have developed a rapid, precise and accurate method for the determinatin of SA and SO in human and animal urine and tissue samples suitable for epidemiological studies and animal experiments. A study carried out on rats fed with diets containing increasing doses of fumonisins, allowed to establish the threshold level of toxins required to trigger the biomarker response in urine and showed a good correlation between the ingestion of fumonisins through the diet and the urinary SA/SO value. The biomarker has been used to evaluate the in vivo effectiveness of cholestyramine and activated carbon in sequestering fumonisins. The addition of cholestyramine to fumonisins contaminated diets was shown to protect rats from the toxic effect of fumonisins. The protective effect of activated carbon was lower than that of cholestyramine. |
