| 内容記述 | [Synopsis] We tried to identify the genes which are involved in the fruiting-body formation of Dictyostelium discoideum. We have selected 17 cDNA clones for the preparation of a knockout construct, from cDNA library of Dictyostelium discoideum slug. They are expressed especially in prestalk cells. The knockout construct was made by insertion of blasticidin S resistance gene into a middle region in cDNAs. As a result, the cells transformed with SSK401 and SSK864 knockout constructs, respectively showed aberrant fruiting bodies and Southern blotting analysis suggested that the knockout constructs were inserted into D. discoideum chromosome DNAs. [摘要] 細胞性粘菌D.discoideumの予定柄細胞特異的に発現するcDNAクローンを用いて、子実体形成に関する遺伝子群の同定を試みた。その結果、ノックアウトコンストラクトSSK401-BsrとSSK864-Bsrの形質転換体は異常な形態形成を示し、またサザンプロット解析によっても染色体DNAへのノックアウトコンストラクトの挿入が確認された。 | |||||||||||||||||
