The steps of the present technique, modified from those of previous techniques based on museum or fresh specimens, are as follows: (1) Soaking of dried fishes for almost a day in water to remove salt before fixation; (2) fixation of specimens in relatively thick formalin; and (3) digestion of muscle with a low-concentration KOH solution.
