High yield of protoplasts was obtained when 5-days cultured mycelia were treated with 0.05 M Na-maleate buffer solution containing 20 mg/ml Cellulase, 20 mg/ml Helicase, 10 mg/ml Zymolyase and 0.5 M MgSO_4 (pH 5.8). Of three strains (TMI 724, 662 and 1247) used, TMI 724 showed the highest yield of protoplasts.Cell wall regeneration of protoplasts was examined by staining with fluorescent brightener (Kayaphor PBS). Percentages of fluorescing protoplasts rapidly increased and reached a maximum 48 hr after treatment with Kayaphor PBS. Protoplasts cultured in a buffer solution showed a fluorescence of Kayaphor but was sensitive to osmotic shock.
