It is generally that the integral membrane plycoprotein with an apparent molecular weight of 80,000 (contact site A) is involved in the cell aggregation process. Contact site A has two kinds of N- glycosidically - linked carbohydrate chains (carbohydrates I and II); carbohydrate I is sulfated and carbohydrate II reacts with wheat germ agglutinin. In this study, we used electrofusion technique to find whether these carbohydrate chains come from different dolichol-linked sugar intermediates and whether these intermediates function in a complementary way to produce native contact site A HL220 mutant cells express contact site A that is 68kd and has carbohydrate I but not II. Cells treated with tunicamycin express contact site A that is 66kd and has carbohydrate II but not I. We fused these two cell types.
