| 内容記述 | [Author abstract]The RAPD-PCR-mediated classification of sweet potato weevils, Cylas formicarius, was performed in order to investigate the geographic variation of their genetic properties in the Southwest islands and Kouchi Prefecture in Japan. When three sets of primers, T13/T13, T05/T05, and T01/T07, were used for PCR, the polymorphic patterns of PCR-amplified DNA were classified into 20 types. The population in each of the tested Southwest islands considered of several types, and type A1B1C3 was common in all tested populations. Moreover, varied DNA patterns were detected in some weevils, suggesting the occurrence of genetic variation in the population. All of the tested weevils which were invaded Kouchi Prefecture in 2000 were type A1B1C1, being commonly detectable in Nakanoshima, Amami, and Okinawa populations. The present study provides useful primer sets for RAPD-PCR, which enabled us to successfully classify sweet potato weevils at the DNA level.[著者抄録]南西諸島のアリモドキゾウムシ個体群のDNAレベルでの地理的分布を調べるため, RAPD-PCR法による識別を行った。 T13/T13, T05/T05, T01/T07の3通りのプライマーでPCRを行い,得られたDNA増幅パターンの多型解析を行った。その結果,アリモドキゾウムシは20タイプに類別され,これらのタイプには南西諸島の全個体群に共通するA1B1C3が認められ,さらにいくつかのタイプの変異型も得られた。また,2000年に高知県で発見された6個体はすべてA1B1C3に類別され,トカラ列島中ノ島,奄美大島,沖縄本島の個体群と同じタイプであることが明らかとなった。 | |||||||||||||||||
