Some of endangered insect species (Red-data listed insects) were detected in our universitycampus, but their survival has been oppressed because of rapid and vigorous propagation of kudzu vine (Pueraria lobata) that caused severe decrease of host plants of insects. In the present study, a basic work was designed to provide biological measures for controlling kudzu for the protection of endangered insects. First, diseased leaves of kudzu were collected to isolate pathogenic bacteria. The candidate isolates obtained were re-inoculated into healthy leaves or stems of the plants to confirm thier pathogenicity to kudzu. Necrotic symptom appeared at inoculation sites 2 days after inoculation, and then expanded to non-inoculated leaves or stems through vessel systems 5 to 6 days after inoculation. Eventually, five pathogenic isolates were selected and identified according to the Bergy's criteria. These bacteria were Rahnella aquatilis MPL-01, Serattia marcescens MPL-02, Pseudomonas cepacia MPL-03, Xanthomonas maltophilia MPL-04 and Pseudomonas sp. MPL-05. The isolated strains were shown to be non-pathogenic to other plant species including solanaceae, cucurbitaceae and leguminosae plants by an artificial inoculation assay. These results suggest that the present isolates would be promising candidates for biological herbicides against kudzu plants. |
