ケモカインは生体内での様々な白血球の移動と局在制御において重要な役割を果たすサイトカインの一群である.本研究ではヒト角膜上皮細胞 (human corneal epithelial cells : HCE) およびヒト角膜実質細胞 (human corneal keratocytes : HCK) でのケモカイン発現能をReverse transcription-polymerase chain reaction (RT-PCR) を用いて網羅的に検討した.両細胞とも,IL-1βやTNF-αなどの刺激により未熟樹状細胞の遊走因子LARC/CCL20,Th1細胞選択的遊走因子MIG/CXCL9,IP-10/CXCL10,I-TAC/CXCL11を,またHCEはTh2細胞選択的遊走因子MDC/CCL22を発現することを見出した.さらに,in vitroでの単純ヘルペスウイルス1型 (herpes simplex virus-1 : HSV-1) 感染はHCEとHCKでLARC/CCL20の発現を,HCEでMIP-1β/CCL4の発現を誘導した.そこで次に,マウスヘルペス性角膜実質炎 (herpetic stromal keratitis : HSK) モデルを用いて,HSV-1感染角膜組織におけるケモカインとケモカイン受容体の発現をRT-PCR解析により検討した.その結果,HSV-1感染後初期からLARC/CCL20,MIG/CXCL9,IP-10/CXCL10の発現が見られ,さらに単球とTh1細胞の選択的遊走因子であるMIP-1α/CCL3,MIP-1β/CCL4,RANTES/CCL5の発現が検出された.それにともなって,それらの受容体であるCCR6,CXCR3とCCR5のシグナルも検出された.これらの発現は病態の改善とともに減少した.一方,Th2細胞選択的遊走因子TARC/CCL17とMDC/CCL22の発現は,病態が改善に向かう後期に検出された.それにともなって,これらの共有受容体CCR4のシグナルが検出された.これらの結果は,HSKの病態はその憎悪期にTh1環境にあり,治癒期にはTh2環境が形成されることを示唆する.このようなマウスHSKモデルにおけるケモカイン発現の経時的変化は,HSKの病態形成過程で観察される経時的な細胞浸潤の変化とよく相関していた.すなわち,HSV-1感染角膜組織におけるケモカイン発現の経時的変化はHSKの病態形成における細胞浸潤機序をよく説明できると考えられた.","subitem_description_language":"ja","subitem_description_type":"Abstract"}]},"item_2_description_36":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"本文データはCiNiiから複製したものである。","subitem_description_type":"Other"}]},"item_2_description_41":{"attribute_name":"フォーマット","attribute_value_mlt":[{"subitem_description":"application/pdf","subitem_description_type":"Other"}]},"item_2_publisher_14":{"attribute_name":"出版者
