c-jun遺伝子は細胞増殖遺伝子として胎生期の関節や関節軟骨の発生に重要な役割を果たしている. また最近c-fos遺伝子が過剰発現すると関節軟骨の変性・破壊につながる可能性も報告されている. 今回, ラット変形性関節症(OA)モデルを作成し, 関節軟骨の変性過程におけるc-fos, c-jun蛋白の発現を免疫組織学的に検索した. その結果, c-fos, c-jun蛋白は, ともにOA誘発の3日目で全層に, 1週目で浅層から中間層に, 2週目で中間層の軟骨網胞核内および細胞質に発現し, 4週目では消失することがわかった. またサフラニンO(S-O)染色による軟骨基質の染色性は, c-fos, c-junの発現にともなって低下した. c-fos, c-lun蛋白の発現がメカニカルストレスの異常に対応してOA発症の初期に軟骨変性に先立ち強く認められたことは, c-fos, c-junによって誘導された軟骨細胞の分裂増殖および幼若化が基質破壊につながる可能性を示唆する.    また, OAにおけるprotein kinase C(PKC)の役割とc-fosとの関係を検討するために, PKC活性化型ホルボールエステルであるphorbol-12-myristate-13-acetate(PMA)を関節内に注入し, 同様にc-fos蛋白の発現を観察した. その結果, PMA注入群ではc-fosの発現が抑制され, S-O染色性の低下は認められず, またOAの進行も明らかに抑制された. これはsecondary messengerであるPKCの活性化がtertiary messengerであるc-fosの発現に抑制的に作用したこと, それにより結果的に軟骨基質が破壊から守られたことを示唆し,
