| 内容記述 | Johns Hopkins大学との国際共同研究により、卵巣高異型度漿液性がんにおける新規免疫調節因子PVRL2の機能を詳細に解析した。ヒト腫瘍検体を用いたシングルセル解析および空間的免疫組織解析により、PVRL2は腫瘍関連マクロファージ（TAM）に特異的に発現し、その発現低下が抗腫瘍性M1様極性を誘導することを見出した。さらに、PVRL2欠損マウスでは、マクロファージの貪食活性が著明に亢進し、T細胞やNK細胞に依存せず腫瘍抑制効果が得られた。これらの成果は、従来の免疫チェックポイント阻害薬が有効性を示さなかったHGSOCに対し、PVRL2を標的とした新たな治療戦略の有望性を示唆するものである。 | |||||||||

| 内容記述 | Through an international collaborative study with Johns Hopkins University, we investigated the function of a novel immunoregulatory molecule, PVRL2, in high-grade serous ovarian cancer (HGSOC). Single-cell and spatial immunohistochemical analyses of human tumor specimens revealed that PVRL2 is specifically expressed in tumor-associated macrophages (TAMs), and its downregulation promotes anti-tumor M1-like polarization. Furthermore, in PVRL2-deficient mice, macrophage phagocytic activity was markedly enhanced, leading to tumor suppression independent of T or NK cells. These findings suggest that targeting PVRL2 may offer a promising therapeutic strategy for HGSOC, which has shown limited response to conventional immune checkpoint inhibitors. | |||||||||
