内容記述 In order to clarify pathological changes in cardiac muscle cell hypertrophy resulting from the development of hypertension, cardiac muscle cells in severely hypertensive M-SHRSP and moderately hypertensive SHR B2 were examined morphologically. The left ventricular myocardial fascicles in these rats consisted of three different layers, superficial, middle and deep. Both the middle and deep layers were thicker in comparison to the superficial layer, and ran spirally, continuing to the apex. Both in M-SHRSP and in SHR B2, there was marked cellular hypertrophy (i.e., accretions in transverse diameter and length) and/or nuclear polyploidization of cardiac muscle cells, particularly in the two thicker layers, according to the increase in blood pressure. As a consequence, concentric hypertrophy was recognized in the left ventricle. Cellular hypertrophy and nuclear polyploidization developed rapidly from an early stage, the latter appearing prior to the former in M-SHRSP. When blood pressure continued to increase above 200 mmHg, cellular hypertrophy development began to weaken with, in particular, little increase in length. During this period, cellular injuries such as intracellular edema and mitochondrial swelling were electron microscopically recognized in the cardiac muscle cells with giant nucleus of 8C or over. These results indicate that hypertensive cardiac muscle cell hypertrophy occurs predominantly in the inner portion of the left ventricle, and is followed by myocardial injury. Since these phenomena were prominent in M-SHRSP at an early stage of life, M-SHRSP seems to be a suitable animal model for studies designed to elucidate pathological changes in hypertrophied muscle cells.
