内容記述 The development of 8 hybrid rat substrains, obtained by backcrossing WT (originally secured by crossing M-SHRSP and WKY) with M-SHRSP and WKY for three to four generations has already been reported. Successive selective brother-sister breeding of these substrains has now been continued for over 20 generations and today 8 inbred substrains have been secured, each with distinctive, different levels of blood pressure, ranging from normal to very severe hypertension. Pathological examination has shown that male M-SHRSP have the most precocious hypertensive vascular changes and that brain and kidney angionecrosis are noted as early as 10 weeks of age while heart, thyroid, adrenal, and testes angionecrosis are found at 11 to 12 weeks of age. Hyaline degeneration of the arterial walls is also seen in the heart, kidney and thyroid from 1 to 2 weeks after angionecrosis begins to develop. Similar changes are also found in these same organs in female M-SHRSP, but in comparison with the males occur 1 to 2 weeks later. In WT-T, the offspring of WT backcrossed with M-SHRSP, earlier and greater incidences of change have been noted as the backcrossings have continued. However, in W-WT, the offspring of WT backcrossed with WKY, no such vascular changes have been seen. These vascular changes begin to appear after blood pressures have surpassed 220 mmHg, but the occurrence of the above pathological symptoms in the various organs depends not only upon the degree and the way in which hypertension develops, but also upon its duration. The incidence of angionecrosis is higher in rats evidencing more rapid increases in blood pressure, whereas the incidence of hyalinization of the arterial wall is greater in rats showing milder elevations. A relatively high incidence of cerebral hemorrhage is noted in rats showing very rapidly developing hypertension, whereas cerebral infarction is more frequently seen in those in which the severe hypertensive state continues over a longer period of time. These findings indicate that because these 8 newly obtained strains present varying degrees of hypertension ranging from mild through severe, as well as because they show a variety of hypertensive vascular changes, they should be useful as experimental animals in detailed studies on human essential hypertension and stroke.
