内容記述 Contractile responses to the receptor-mediated agents, noradrenaline, phenylephrine, serotonin and angiotensin II, and the voltage-dependent agents, such as K+ and Ca2+, were investigated on the vascular strips of the aorta, the superior mesenteric, the femoral and the common carotid arteries of 3,6,12 week-old malignant stroke-prone SHR (M-SHRSP). The results were compared to those of the age-matched SHRSP and WKY as the control. At 3 weeks of age, the magnitude of the contractile responses to these agents were in the order of M-SHRSP>SHRSP≧WKY in the blood vessels tested. At 6 weeks of age, the magnitude of the contractile responses to these agents were in the order of M-SHRSP>SHRSP>WKY in all of the blood vessels tested. The contractile responses to the receptor-mediated agents, noradrenaline, phenylephrine, serotonin and angiotensin II of 12 week-old M-SHRSP were smaller than those of aorta and the common carotid artery in 6 week-old SHRSP. The contractile responses to the receptor-mediated agents of 12 week-old SHRSP were larger than those of 6 week-old SHRSP in all of blood vessels tested, while the responses of 12 week-old WKY were similar or slightly larger than those of 6 week-old WKY. This difference is depending on the kind of blood vessels tested. Thus, the contractile responses to the receptor-mediated agents in 12 weeks of age were in the order of SHRSP>WKY>M-SHRSP. However the contractile responses to the voltage-dependent agents, K+ and Ca2+, were in the order of M-SHRSP>SHRSP>WKY in all blood vessels in 12 weeks preparation. The receptor affinities (sensitivity of receptor) in the blood vessels of 3,6 and 12 week-old M-SHRSP were not significantly different from those of age-matched SHRSP and WKY. These results suggest that the increased contractile responses to the receptor-mediated agents are affected by the elevation of blood pressure, and that the extremely high blood pressure affects the number of receptors, though the receptor affinity was not affected by high blood pressure.
