In bacteria, in vitro repair of thymine dimers can be achieved by the sequential action of a Micrococcus luteus endonuclease and DNA polymerase I. The repair activity of DNA polymerase I is associated with its 5'→3' exonucleolytic activity. DNA polymerase activities were purified from rat liver. Partially purified enzyme was passed through a DNA cellulose column. The DNA has previously been nicked with a repair endonuclease purified from rat liver. Three fractions were obtained. The fractions were then eluted on a SDS Sephadex G200 column. The first fraction (170,000 daltons) has only DNA polymerase activity, the second (40,000 daltons) has both 5'→3' exonuclease and DNA polymerase activity. The third fraction (18,000 to 20,000 daltons) has only exonucleolytic activity. The second fraction was recovered with both polymerase and exonuclease activities. |
