内容記述 Unilateral transection of the sciatic nerve in the frog resulted in a decrease in the relative wet weight of the sartorius muscle about 4 weeks postoperatively in contrast to the contralateral side. Energy metabolism was compared between the denervated and the contralateral innervated muscles at rest and during contracture with 31P nuclear magnetic resonance (NMR). The denervated muscle showed a decrease in the relative concentration of phosphocreatine (PCr) to β-adenosine triphosphate (β-ATP) at rest, which started about 4 weeks after denervation and this state indicated a decrease in the energy store. Sartorius muscles, 3-6 weeks after denervation, showing a wet weight decrease of 95-75% were perfused with a potassium solution to induce potassium contracture for determination of serial changes in the PCr/β-ATP ratio. The contraction-associated decrease in PCr/β-ATP and an increase in inorganic phosphate (Pi)/β-ATP of the denervated muscle were less marked than those in the control. In caffeine contractures, the denervated and innervated muscles showed similar serial changes in the PCr/β-ATP and Pi/β-ATP ratios. After a lapse of 1 week or more following denervation, the muscular tension induced by potassium contracture decreased, but no decrease was observed in the caffeine contracture, because of the inhibition of excitation-contraction coupling. It is suggested that phosphate metabolism of the denervated muscles during contracture is parallel to the change of tension, showing little difference from the innervated muscles in the early stage of atrophy.
