| 内容記述 | It has been reported that the spontaneously hypertensive rat (SHR) is a useful animal model for the study of human essentilal hypertension. It is generally accepted that blood vessel tone depends on the contractile and relaxation responses. Thus, a reduced relaxation response might induce elevated blood pressure. Rat tail is an important organ for the body balance and heat radiation. There are very few reports concerning the effects of blood pressure on the responses to the relaxants in the tail artery. In the present study, the relaxation responses of taii arteries from stroke-prone SHR (SHRSP) and Wistar-Kyoto rats (WKY) were investigated from the standpoint of aging. Some of experimental results were compared to those of the aorta: Relaxation responses to acetylcholine (ACh) and histamine, endothelium-derived relaxing factors decreased with age in both SHRSP and WKY tail arteries, with SHRSP exhibiting lower values. The same results were obtained in SHRSP and WKY aortas. The relaxation responses to ACh and histamine in the tail artery were less than in the aorta. Examination of the relaxation responses to nitroprusside and nitroglycerin revealed that NO radical-containing drugs did not alter with age in both SHRSP and WKY tail arteris, and were not different between SHRSP and WKY tail arteries. The same results were obtained in SHRSP and WKY aortas. The relaxation responses to nitroprusside in tail arteries were somewhat less than those in aorta. The relaxation response to isoproterenol (Iso), a β-receptor stimulating drug decreased with age in both SHRSP and WKY tail arteries, but to a great degree among SHRSP. The relaxation responses to Iso almost disappeared in 6-month-old SHRSP. The effects of forskolin and dibutyryl cAMP did not change with age in both SHRSP and WKY tail arteries. Also, the relaxation responses to those drugs were not different between SHRSP and WKY. The relaxation responses to diltiazem, a Ca2+ antagonist, in tail arteries were reduced in WKY, not in SHRSP. | |||||||||
