内容記述 Angiotensinogen, the protein precursor of angiotensin II which is a critical regulator of blood pressure and electrolyte balance, is constitutively produced by the liver and secreted into the circulation. The 5'-flanking region of the human angiotensinogen gene has been found to regulate its own expression in the liver of transgenic mice, but the transcriptional regulation of the human angiotensinogen gene has not been reported. We analyzed its promoter region by utilizing the reporter plasmids that contain the 5'-flanking region with various lengths fused to the bacterial chloramphenicol acetyltransferase (CAT) gene. In transiently transfected human hepatoma HepG2 cells, the only 32-bp DNA region located at nucleotide positions between -344 and -313 bp was shown to mediate half as much as the full extent of parental promoter activity. An electrophoretic mobility shift assay (EMSA) using this segment identified the formation of DNA/HepG2 nuclear factor complexes. These complexes could be dissociated by competing recognition sequences that contain those of either Sp1, a widely expressed GC box-binding factor, or RBF-1, a recently characterized GC box-binding factor regulating the human retinoblastoma (Rb) gene. An in vivo competition experiment also suggested that the CAT activity derived from the cotransfected angiotensinogen promoter is reduced by a competitor containing RBF-1 binding sites or containing Sp1 binding sites. These findings suggest that Sp1 and RBF-1 play an important role in maintaining the constitutive promoter activity of the human angiotensinogen gene and that the angiotensinogen gene is characterized as the second target for RBF-1 beside the Rb gene.
