| 内容記述 | Chronic myelogenous leukemia is characterized by the Philadelphia chromosome, which results from the fusion of the bcr gene and the abl gene. The bcr-abl fusion gene encodes a tyrosine kinase with transforming activity. Inhibition of bcr-abl gene expression may have profound effects on the cell biology of Ph positive cells and become a therapeutic technique. In this study, a synthesized ribozyme targeted to the b3a2 fusion gene was used. The ribozyme cleaved bcr-abl mRNA in cell free in vitro system. Then, this ribozyme was introduced into a murine myeloproliferative sarcome virusbased retroviral vector. The recombinant retrovirus was infected to CML cell line, K562. The ribozyme decreased levels of bcr-abl mRNA in these cells, and inhibited cell growth partially. These results suggest that the ribozyme could be an effective therapeutic agent for the treatment of Ph positive leukemias. | |||||||
