| 内容記述 | Arbekacin (ABK) and vancomycin (VCM) are used for treating infections caused by methicillin-resistant Staphylococcus aureus. A rapid, accurate, coupled column chromatographic procedure employing direct injection of microserum has been developed for ABK and VCM quantitation in human serum. The columns consist of a separation column filled with reversed ODS-C18 and a pretreatment column filled with macroporous hydrophilic copolymer. The procedure for ABK determination involves the counter-ion method with sodium octanesulfonate, subsequent derivatization with o-phthalaldehyde and fluorescence detection. A mixture of potassium phosphate buffer and acetonitrile is used as the mobile phase, and VCM is directly detected at a UV wavelength. Coefficients of variation for within-day reproducibility were 0-3.00% (x̄=1.2-21.1 μg/ml) and 1.08-2.88% (x̄=2.6-52.4 μg/ml) for quantitating ABK and VCM, respectively ; for between-day reproducibility they were 4.68% (x̄=2.5 μg/ml) and 3.47% (x̄=9.5 μg/ml), respectively, with recovery rates of 92-106% and 98-112%, respectively. The minimum limit of detection (S/N=3) was 0.02 μg/ml for the ABK assay and 0.05 μg/ml for the VCM assay. The correlation coefficients between the fluorescence polarization immunoassay and this method were 0.978 and 0.991 for ABK and VCM, respectively. No interference was found either from endogenous substances or from many currently used antibiotics, indicating good selectivity and high sensitivity. | |||||||||
