内容記述 Since blood vessel tone depends on the contractile and relaxation responses to the agents, the decreased relaxation response might induce hypertension. In this study, I examined the responses to many relaxants in malignant stroke-prone spontaneously hypertensive rat (M-SHRSP) to clarify the relation-ship between the onset and maintenance of hypertension and the relaxation responses of blood vessels. The findings obtained were compared with those obtained in the age-matched Wistar Kyoto rats (WKY). The relaxation responses to high concentrations of ACh or histamine, endothelium-derived relaxation factor (EDRF) releasing drug, were weaker in M-SHRSP than in WKY in the superior mesenteric artery, but those to in the low concentrations were not different between M-SHRSP and WKY. However the relaxation responses to ACh or histamine in the aorta were not different between M-SHRSP and WKY in the concentrations used. The relaxation responses to the high concentrations of ACh in the superior mesenteric artery were increased by the treatment with indomethacin, but those in the M-SHRSP and WKY aortas and the WKY superior mesenteric artery were not affected by this treatment. The relaxation responses to nitroprusside in the aorta and the superior mesenteric artery were not different between M-SHRSP and WKY. The relaxation responses to high concentrations of isoproterenol in the superior mesenteric artery of M-SHRSP were weaker than those of WKY, but those in the aorta were not different between M-SHRSP and WKY. The relaxation responses to aminophylline, forskolin, dibutyryl cyclic mono-phosphate and diltiazem in the aorta and the superior mesenteric artery were not different between M-SHRSP and WKY. The following conclusions were made. 1) The decreased effects of EDRF-releasing drugs and of β-stimulant start at the smaller artery compared to larger artery, which may be related to the onset of hypertension of M-SHRSP. 2) The reduced relaxation responses to ACh or histamine is due to the increased secretion of endothelium-derived contracting factor, but is not due to the decreased secretion of endothelium-derived relaxing factor. 3) The reduced relaxation response to ACh or histamine is not due to the decreased activity of guanylate cyclase. 4) The decreased effect of isoproterenol is not due to the decreased activity of adenylate cyclase or the increased phosphodiesterase activity, but is due to the decreased sensitivity of β-receptor.
