内容記述 The effects of inosine, guanosine 5'-monophosphate (GMP), cytidine, uridine, thymidine, and their mixture (4 : 4 : 4 : 3 : 1 in molar ratio, OG-VI) on cell growth were determined by examining the DNA and RNA syntheses of the primary monolayer culture of hepatocytes and a hepatoma cell line, AH130,in vitro, liver regeneration after hepatectomy and malignant tumor growth in vivo. The in vitro findings suggested that addition of optimal amounts of nucleosides and nucleotides enhanced the growth of hepatocytes and hepatoma cells, although the optimal concentration was about 10 times higher in hepatoma cells than in cultured hepatocytes. The nucleoside and nucleotides mixture of OG-VI enhanced the DNA and RNA syntheses in the primary monolayer culture of hepatocytes, and inhibited the syntheses of DNA and RNA by hepatoma cells at a relatively high concentration. In vivo studies revealed that the OG-VI mixture was well captured in various organs, including the liver, and that the preoperative administration of the OG-VI mixture facilitated the liver regeneration after partial hepatectomy. More-over, the OG-VI mixture did not enhance tumor growth in vivo when administered within the range of concentrations which stimulate liver regeneration.
