The antinociceptive effects of intrathecal (i.t.) administration of L-arginine were investigated in rats. When assessed with the tail-flick test, i.t. administration of 1000 μg L-arginine had significant antinociceptive effects in rats. At a lower dose (100 μg), L-arginine failed to suppress the nociceptive response. When similarly treated, however, D-arginine (1000 μg) failed to suppress the nociceptive response. Furthermore, suppressive effects with 1000 μg of L-arginine were significantly reversed by naloxone (10 μg ; i.t.) pretreatment. It is, therefore, speculated that L-arginine-induced suppression of nociception in the spinal cord is mediated through the activation of opioid receptors. These findings suggest that L-arginine plays a important role in processing nociceptive inputs in the spinal cord. |
