内容記述 For use in repair of articular cartilage defects, autografts were prepared by embedding and culturing cultured costal chondrocytes in collagen gels. We collected the outgrown chondrocytes from autogenous costal cartilage primarily cultured in a monolayer for 7 days to acquire a large number of cells. These cultured, costal chondrocytes were embedded into collagen gels and were further cultured for 10 days to acquire the appropriate hardness for use as autografts. In these autografts, the cell number increased two-fold and the cells incorporated 35S. At 18 days after the first surgery, autograft transplants were sized to fit defects, 5 mm in diameter, in rabbits extending to the depth of the subchondral bone in the patellar groove. Histological examination revealed that for 30 weeks after transplantation, the defects were covered with a hyaline-like cartilage. Biochemically, the repaired tissues possessed Type II collagen. Therefore, our results show that costal chondrocytes can be used for the purpose of repairing articular cartilage defects.
