内容記述 The tibiae of 15 young rabbits were osteotomized and lengthened gradually to produce a gap of 1 cm over a period of 10 days, and then one dose of 0.6 mg/kg of bisphosphonate (pamidronate disodium) was injected intravenously into 5 of them. The bone mineral density (BMD) in the proximal and distal portions of the tibia and lengthened callus was evaluated using dual energy X-ray absorptiometry every 5 days for 30 days after lengthening had been completed. In the control rabbits the BMD in the distal and proximal diaphyseal portions of the tibiae continuously decreased to about 50% 15 days after completion of lengthening. In the treated rabbits, however, the BMD increased until day 10 in the proximal portion and until day 5 in the distal portion, with the difference being significant in the proximal portion. It was demonstrated that a single pamidronate dose could inhibit bone resorption in an area of high bone turnover. The stimulatory effect of a single dose on the mineralization of the lengthened callus, however, was not significant. Experiments using repetitive doses of pamidronate for longer periods should be mandatory.
