内容記述 Hyaluronic acid (HA) is known to be present in wounded corneal tissue and in the tear fluid and to play an important role in corneal wound healing. However, the origin of HA on the ocular surface is still unclear. In this study, HA localization after corneal epithelial debridement and synthesis of HA by cultured corneal epithelial cells were investigated. The effect of wounding of cultured corneal epithelial cells on synthesis of HA was also investigated. The central rabbit corneal epithelium (10mm in diameter) was scraped off and a tissue section made after three days was stained with biotinylated HA binding protein (HABP) and the avidin-biotin-peroxidase complex method. Cultured rabbit corneal epithelial cells were used for the measurement of HA in the culture medium and in the cell layer. The amount of HA was measured with HABP using the HA rapid assay kit. The amount of HA in the culture medium two days after scratching the surface of the confluent cultured corneal epithelial cells with 23G needles was also measured. It was demonstrated that specific HA staining was observed underneath and in the intercellular space of migrated rabbit corneal epithelial cells three days after the debridement. The amount of HA both in the culture medium and in the cell layer increased significantly in proportion to the number of cells. The amount of HA in the culture medium was about seven times higher than that in the cell layer. The amount of HA in the culture medium increased with the cultivation periods up to seven days, but that in the cell layer did not change. The amount of HA in the culture medium increased significantly (p<0.01) in proportion to the number of scratching wounds on the confluent cultured corneal epithelial cells in vitro. These findings demonstrated that cultured corneal epithelial cells had the ability to synthesize HA and the most of them were released into the culture medium. In the wounded condition, synthesis of HA by cultured corneal epithelial cells was stimulated. It was suggested that corneal epithelial cells synthesized HA and a part of HA which accumulated in the wounded cornea was produced by migrating corneal epithelial cells in vivo.
