内容記述 The purpose of this experiment was to clarify morphologically the differences in alveolar damage due to NO2 inhalation between young and old rats. Three- and 85-week-old rats were used in the experiment. The animals were exposed to 15 ppm NO2 continuously for 18 days. The lung tissue was examined chronologically by both light and transmission electron microscope. In old rats, alveolar edema was observed clearly 4 days after initial exposure. At seven days after exposure, the alveolar edema had disappeared, but marked interstitial edema was observed in the vas circumference. In young rats, neither alveolar edema nor interstitial edema were observed during the exposure period. Observation under a transmission electron microscope of old rats 4 days after exposure, showed desquamation from the basal membrane in the alveolar 1 type epithelium, and swelling and vacuolation in the capillary endothelium. In old rats 7 days after exposure, the capillary endothelium showed a decrease in the pinocytotic vesicles and disruption of the tight junction. Desquamation from the basal membrane was not observed in the alveolar 1 type epithelium. In old rats 18 days after exposure, the capillary endothelium showed a decrease in pinocytotic vesicles and bleb formation. The endothelial and epithelial damage was slighter in young rats than in old rats during the exposure period. Our observation has clarified that endothelial damage continued during exposure, and that endothelial cells are more sensitive to NO2 damage in aged rats, resulting in edema and interstitial fibrosis.
