We evaluated the detection of ABH antigens in paraformaldehyde-fixed human cardiac muscle by the EnVision+ system, which employs a dextran polymer reagent for the secondary antibody. Immunohistochemical microscopy of blood group A and B tissues using monoclonal anti-A and anti-B showed strong staining of the cardiac interstitium, but muscle fiber itself was negative. There was also extensive staining of the endothelial cells lining blood vessels. Cardiac muscle fibers were strongly stained by anti-H, but endothelial cells and smooth muscle of blood vessels were stained weakly and the interstitium was negative. Both endothelial cells lining blood vessels and the cardiac interstitium of blood group O tissues were strongly stained by anti-H, and cardiac muscle itself was stained weakly. In highly putrefactive cardiac muscle samples incubated at 37℃ in airtight tubes for one month, ABO blood group could be determined by the EnVision+ system. |
