Alveolar macrophages (AM) in BALF were found to contain urokinase-type PA (u-PA), tissue-type PA (t-PA), or both types of PAs. When AM were cultured, AM secreted only u-PA into the culture medium, and the cell extract contained u-PA only. The influences of various pharmacological agents on PA secretion by cultured AM were investigated ; sulpyrine and hydrocortisone reduced PA secretion in culture medium, but OK-432, histamine, interferon-γ (IFN-γ) and platelet activating factor (PAF) increased PA activity in culture medium. PA activities in culture medium and cell extracts were decreased by the addition of actinomycin D, cycloheximide, and 5-fluorodeoxyuridine. In order to clarify the mechanism of secretion of u-PA by cultured AM, a model cell, U-937, a human monocyte-macrophage, was used. The secretion and synthesis of u-PA by U-937 were accelerated by PAF, IFN-γ or OK-432.
