内容記述 The interaction of eosinophils and the bronchoalveolar lavage (BAL) cells was studied employing an animal model of the late asthmatic response (LAR) in the guinea pig, characterized by the bronchial eosinophilia at the time of LAR. Hartley-strain male guinea pigs each weighing 200-250 g were treated with 30 mg/kg cycclophosphamide intraperitoneally, and two days later were sensitized primarily with intraperitoneal injection of 10 or 1,000 μg ovalbumin (OA) and 100 mg Al(OH)_3, and 3 weeks later were given a booster injection of 10 μg OA and 100 mg Al(OH)_3. Four weeks later, BAL was performed, and parts of the BAL cells were cultured with and without OA stimulation for 24 hours. The effects of the BAL cell culture supernatant on the activity of the human eosinophilic cell line (EoL-3) and eosinophils isolated from normal human peripheral blood were examined. Furthermore, the function of BAL cells altered by sensitization was also assessed. 1) Pretreatment of eosinophils with the culture supernatant of BAL cells in the OA sensitized groups enhanced the luminol dependent chemiluminescnse (LDCL) to calcium-ionophore A23187, and the enhancement of their peak values followed increasing OA dosage administered for sensitization. 2) Incubation of eosinophils with the culture supernatant in the sensitized groups resulted in the augmentation of cytotoxicity for bronchial epithelial cells (AK-D). 3) Eosinophil chemotactic activity of culture supernatants was enhanced in the sensitized groups. 4) BAL cell-mediated LDCL to A23187 was increased in the OA sensitized groups. 5) BAL cell-mediated cytotoxicity for AK-D was not altered by OA sensitization. These observations suggest a potential role for not only eosinophils but also BAL cells in this model in the bronchial inflammation that may be relevant to the pathogenesis of LAR.
