| 内容記述 | The regulatory mechanism of Interleukin-6 (IL-6) production in cultured human keratinocytes in the presence of the Transforming growth factor (TGF)-α and β1 was examined by determining the amounts of both IL-6 messenger RNA (mRNA) and protein. Normal human epidermal keratinocytes and cells of human trichilemmoma cell line were used. Proliferation of normal human epidermal keratinocytes was stimulated by TGF-α, but inhibited by TGF-β1. Proliferation of cells of human trichilemmoma cell line was not affected by either TGF-α or TGF-β1. In the Northern blot analysis with IL-6 complementary DNA probe, expression of IL-6 mRNA in cells of human trichilemmoma cell line reached the peak 24-48 h after addition of both TGF-α, β1 in the presence of 10% fetal calf serum (FCS). However, in the presence of 1% FCS, the plateau of IL-6 mRNA shifted to an early phase by the addition of TGF-β1. In the enzyme labeled immunosorbent assay with IL-6 monoclonal antibody, IL-6 secretion by cells of human trichilemmoma cell line was upregulated in the presence of TGF-α and β1. These findings indicate that TGF-α and β1 upregulate the expression of IL-6 mRNA and protein. We assume that a cytokine network exists in the epidermis and participates in cell proliferation, wound healing and various other biological responses. | |||||||||
