| 内容記述 | The effects of intrasplenic injection of interleukin-2 (IS-IL-2) on the lymphokine-activated killer (LAK) activity in the liver/spleen and the migration of the LAK cells generated in the spleen were examined in C3H/He mice. LAK activity of the liver lymphocytes from IS-IL-2 injected mice resulted in potent LAK activity. Histological findings revealed that large blastic lymphoid cells significantly increased more than 20% by IS-IL-2 injection. The γδT cell receptor positive cells, considered as one of the compartments of anti-tumor effector system, also increased by IS-IL-2 injection detected by FACS analysis. These results may be reflected our previous findings in beagle dogs that the intrasplenic arterial infusion of IL-2 results in the high concentration of IL-2 in the portal vein and high concentration IL-2 directly activate the infiltrating lymphocytes or the endothelial cells in the liver. We next examined whether the LAK cells in the spleen migrate to the liver. ^<51>Cr-labeled LAK cells injected intrasplenically and followed the distribution of the radioactivity of labeled cells throughout each organs. The results demonstrated the majority of the count were already in the liver within 3hours and were retained in the liver for at least 24hours. To assess the in vivo efficacy C3H/He mice having liver metastases were treated IS-IL-2 injection daily for 3days and followed the survival time. Survival rate definitely improved and some mice were cured. These finding suggest that the regional administration of IL-2 to the spleen can efficiently induce LAK effectors capable of migrating to the liver. Such a strategy which employs regional infusion of IL-2 to the spleen may be applicable for the treatment of hepatic metastases in humans. | |||||||||||||
