| 内容記述 | Membrane fluidity and phospholipid composition of sinusoidal membrane vesicles [SMV] and canalicular membrane vesicles [CMV] of rat hepatocytes were studied to investigate their relevance to the topological difference in membrane function. Membrane fluidity was measured by fluorescence polarization of 1,6-diphenyl-1,3,5-hexatrien [DPH] and by the spin label method using 5-doxyl stearic acid [5-DSA] and 16-doxyl stearic acid [16-DSA] as probes. Fluidity of CMV was significantly lower than that of SMV in the both methods. CMV had a significantly higher total phospholipid content than SMV. Phospholipid composition was determined by a newly developed high performance liquid chromatography [HPLC]. The level of sphingomyelin was significantly higher and the level of phosphatidylcholine lower in CMV than SMV. The cholesterol level was significantly higher in CMV than in SMV. These alterations of lipid composition in CMV are considered to be related to the decrease in fluidity. Addition of S-adenosyl-L-methionine [SAMe], which is reported to improve cholestasis, to SMV resulted in an increase in fluidity and Na^+, K^+-ATPase activity. These changes may contribute to the choleretic effect of SAMe. | |||||||||
