内容記述 | To elucidate the membrane abnormality in hypertrophied SHR myocardium, the sarcolemmal permeability was morphologically examined using horseradish peroxidase (HRP). Doxorubicin was administered to 16-week-old male SHR and age-matched WKY and hearts were examined at the 1st and the 4th day after the administration. In SHR myocardium, HRP were found in the papillary muscle and a part of the subendcardial myocytes on the 1st day, and recognized in many myocytes throughout the entire left ventricular wall on the 4th day. Electron microscopic examination revealed that HRP were located in 1-, t-tubes or intercalated disc. Moreover, abundunt HRP were also seen along with myofilaments in markedly degenerated myocytes. However, few HRP were seen in either controls or treated WKY myocardium on either light or electron microscopy. These findings clearly demonstrate the marked increase in membrane permeability in SHR myocardium at the early stage of membrane change in doxorubicin cardiomyopathy and supports our previous biochemical studies on the membrane vulnerability of SHR heart which may be caused by lipid peroxidation. | |||||||||||||||||
