Plasminogen activator inhibitor (PAI) was purified from the renal tissues of spontaneously hypertensive rat-stroke prone (SHRSP) and that of Wistar Kyoto rat (WKY) using salting-out, acidification, and chromatography with anti-PAI-1 IgG-Sepharose and DEAE cellulose column. The purified PAI from both rats showed a single band with a molecular weight of 42 kDa in SDS-PAGE and reverse fibrin autography. The purified PAI inhibited human tcu-PA, sct-PA and tct-PA by the synthetic substrate S-2251 method, but did not act toward scu-PA. The PAI activity of the homogenized solution of the kidney of WKY was 1.2 times as high as that of SHRSP. These findings suggest that a quantitative difference in the PAI activity in the kidney of SHRSP and WKY is involved in the renal failure of SHRSP. |
