内容記述 The localization of α_1-adrenergic receptors in rat renal glomeruli was examined by using the radioligand receptor assay and patch clamp technique. In Sprague-Dawley (SD) rats, there were two different kinds of glomerular α_1-adrenergic receptors, one had high affinity and low binding capacity and another had low affinity and high binding capacity. In the rats exposed to cold stress, glomerular α_1-adrenergic receptors decreased significantly, while the urinary excretion of catecholamines markedly increased. Wister Kyoto rats (WKY) had two distinct affinities of glomerular α_1-adrenergic receptors and did not show any significant changes between the ages of 6 and 10 weeks. On the other hand, the M strain of stroke-prone spontaneously hypertensive rats (M-SHRSP) had only one type of glomerular α_1-adrenergic receptors. However, these affinities in M-SHRSP were quite different between the two ages. For determining the localization of α_1-adrenergic receptors in glomeruli, mesangial cells were cultured from the glomeruli of SD rats. Cultured rat mesangial cells were contracted by phenylephrine, and this contraction was attenuated by the α_1-blocker bunazosin. In the patch clamp experiment, two different conductances of K^+ channels were observed from cultured rat mesangial cells activated by phenylephrine under conditions of inside-out patch. These channels were blocked by Ba^<2+>, which implys that these carriers were K^+ ions. These findings suggest that α_1-adrenergic receptors exist in rat glomeruli and that they are localized in the mesangial cell.
