内容記述 | Thyroid cells obtained from 21 normal human thyroid tissues and 17 diseased thyroid tissues (1 Graves' disease, 4 follicular adenomas, 11 papillary carcinomas and 1 follicular carcinoma) were cultured in collagen gel, and then, morphological characteristics and thyroglobulin (Tg), triiodothyronine (T_3) and thyroxine (T_4) products were examined. In this study, the results were obtained as follows. Follicular adenoma cells and papillary carcinoma cells of the thyroid in collagen gel culture could survive for a longer term than in monolayer culture. In collagen gel culture, colonies revealed a three dimensional in vivo-like structure and demonstrated characteristic morphology depending on the orignal structure of the thyroid. In immunohistochemical analysis, Tg production was observed in colonies formed from 6 normal thyroid, 1 Graves' disease, 4 follicular adenomas, 8 papillary and 1 follicular carcinomas. T_3 production was observed in the colonies produced by 2 papillary and 1 follicular carcinomas. T_4 production was also observed in the colonies of 3 papillary and 1 follicular carcinomas. After addition of thyroid stimulating hormone (TSH) to medium in collagen gel culture, the colonies produced by normal thyroid gland cells showed differentiation, such as enlarged follicle and columnar epithelium. However there were no morphological changes in the colonies produced from diseased thyroid tissue. These findings suggested that collagen gel culture offered an in vivo-like environment to cultured cells and were a useful method for survival and differentiation of thyroid cells. | |||||||||
