内容記述 | To determine whether activation of the K-ras gene, and allelic deletions on chromosomes 17p and 18q play an important role in the progression of colorectal carcinoma, we analyzed DNAs from 100 pairs of colorectal carcinoma and matched normal colonic mucosal tissues from the same patients. Slot blot analysis for mutation of K-ras at codon 12 showed that 42% of colorectal carcinoma DNAs possessed an activated K-ras gene. Although no correlation of the frequency of activation of K-ras gene to the histopathologic stage in colorectal carcinoma was observed, the frequency of K-ras activation in stage V carcinoma was higher than that in stage I carcinoma. Southern blot analysis for loss of heterozygosity on chromosomes 17p and 18q demonstrated that DNAs from 35 out of 56 informative patients (63%) and 30 out of 49 informative patients (61%), respectively, had deletion of one allele on chromosomes 17p and 18q. In addition, association between allelic deletions on chromosomes 17p and 18q was observed. The incidence of allelic deletion on chromosome 17p in colorectal carcinoma with distant metastasis was significantly higher than that in carcinoma without distant metastasis. These findings indicate that genetic alterations including activation of K-ras gene and allelic deletions on chromosomes 17p and 18q play an important role not only in the early stage but also in the late stage of colorectal carcinoma development. Furthermore, examination for allelic deletions on chromosomes 17p and 18q may provide a useful tool for predicting prognosis of the patients with colorectal carcinoma. | |||||||||||||
