内容記述 | The development of biotechnology has resulted in successful gene cloning of cytokines, and elucidation of the regulation of immunocompetent cells. However, the function of each cytokine on individuals with cancer remains unknown. Interleukin 1β (IL-1β), tumor necrosis factor α (TNFα) and interleukin 6 (IL-6) are mainly produced by a monocyte line. These three cytokines overlap with many functions. In this study, the in vitro IL-1β, TNFα and IL-6 production stimulated with lipopolysaccharide (LPS) were examined in human macrophages (Mφ) obtained from healthy subjects and patients with cancer of the upper digestive tract (gastric and esophageal cancer) to elucidate the influence of these cytokines on the cancer patient. Mφ were isolated from the peripheral blood using the Ficoll-Conray centrifugation technique and subsequently with the plastic disc adherence technique. They were incubated at 37℃ in 5% CO_2 air with or without LPS. IL-1β and TNFα released into the culture fluid were measured by immunoradiometric assay (IRMA), and IL-6 was measured by enzyme-linked immunosorbent assay (ELISA). The production of these three cytokines were detectable within 4 hours of incubation, however, at 2 hours, only TNFα was detected in the culture supernatant obtained from healthy subjects and cancer patients, thus presumably affecting the production of IL-1β and IL-6 in the culture. The in vitro producibility of IL-1β by Mφ was found to be significantly enhanced in patients with esophageal cancer and gastric cancer as compared with that in healthy subjects (p<0.05, p<0.005). Similarly, the in vitro producibility of TNFα and IL-6 by Mφ was also found to be significantly enhanced in patients with esophageal cancer and gastric cancer as compared with that in healthy subjects (p<0.05, p<0.0005), (p<0.0001, p<0.0005). A positive relationship among all three cytokines was recognized in the healthy subjects, while a positive correlation was recognized only between IL-1β and TNFα in patients with gastric cancer. The production of IL-1β by Mφ in gastric cancers was found to be significantly enhanced in patients with stages III and IV cancer as compared with that in patients with stages I and II cancer. The production of IL-1β in patients with ps(+) and Iy(+) was significantly increased as compared with that in patients with ps(-) and ly(-) clinicopathological factors. The production of TNFα in patients with ps(+) significantly increased as compared with that in ps(-). These findings suggest that measurement of the production of three cytokines by Mφ can be used to estimate the immunological status of patients with cancer. | |||||||||
