| 内容記述 | To investigate the association between amplification of the c-erbB-2 gene and overexpression of the c-erbB-2 oncoprotein, we studied prospectively a series of 90 primary breast cancer patients, using the Southern blot method and immunohistochemical staining. Amplification of the c-erbB-2 gene was detected in 25 of the 90 tumor samples (28%), and overexpression was also found in the same frequency. Although tumors with overexpression of the c-erbB-2 oncoprotein did not always show the gene amplification, there was a strong correlation of amplification of the c-erbB-2 gene with overexpression of the c-erbB-2 oncoprotein. These findings demonstrate that immunohistochemical staining of the c-erbB-2 oncoprotein is useful for analyzing c-erbB-2 gene alteration in breast cancer. Then, we analyzed for overexpression of the c-erbB-2 oncoprotein in 263 breast cancer between 1985 and 1990 by immunohistochemical techniques, to determine whether overexpression of the c-erbB-2 oncoprotein can be used to predict the prognosis of the breast cancer patients. Overexpression was found in 19% of the 263 tumor samples. The tumor with overexpression of the c-erbB-2 oncoprotein showed poor prognosis in both overall survival and disease free survival, compared with the tumor without overexpression. Multivariate analysis indicated that overexpression of the c-erbB-2 oncoprotein is an independent predictor of the patients outcome. Therefore, immunohistochemical analysis of the c-erbB-2 oncoprotein may be helpful in characterizing new biological subclasses in breast cancer. | |||||||||||||
