Osteoblast-like cells (HT-3) have protein kinase C and that alkaline phosphatase activity is reduced with activation of protein kinase C. However, alkaline phosphatase activity is also regulated by prostaglandin E2, since this activity is reduced in the presence of this prostaglandin. Since it has been shown clinically that induced micromovement leads to rapid healing of long bone fractures, we investigated the effect of applied cyclic stretch on the alkaline phosphatase activity of HT-3 cells, using a computerized and pressure-operated instrument that physically deforms the cells. As cyclic stretch enhanced alkaline phosphatase activity in the cells, the micromovement seems to have a direct effect on osteoblasts. Cyclic stretch reduced production of prostaglandin E2.
