内容記述 | The non-obese diabetic (NOD) mouse, which was isolated from the ICR mouse, is an animal model of human insulin-dependent diabetes mellitus (IDDM). Although many cell species including T-cells, natural killer (NK) cells and macrophages have been implicated in the pathogenesis of IDDM, there are few reports concerning the role of B-cells in the disease. In this study, the number of splenic IgG- and IgM-secreting cells was determined in female NOD mice by the plaque forming cell (PFC) assay, and compared with the results obtained from ICR mice. In addition, in view of the fact that cyclophosphamide (CY) can promote the onset of diabetes in NOD mice but not in ICR mice, the effects of CY on splenic cells were evaluated in female ICR and female NOD mice by the PFC assay and flow cytometric analysis using monoclonal antibodies. All the levels of IgG-PFC, IgM-PFC, and serum immunoglobulins were higher in NOD mice than in ICR mice. There was no difference in the number of IgG- or IgM-PFC between NOD mice with or without diabetes. The number of IgG- and IgM-PFC were both higher in the CY-treated NOD mice than in the saline-treated NOD mice. However, the ICR mice did not show any significant difference in the number of IgG- or IgM-PFC between CY-treated and saline-treated groups. Similar findings were obtained in flow cytometric analysis using LR-1, a monoclonal antibody specific to murine B-cells : LR-1 positive cells were increased after CY-treatment in NOD mice but not in ICR mice. These findings indicate that polyclonal activation of B-cells is intrinsic in NOD mice and this characteristic is further enhanced by CY-treatment. | |||||||||
