内容記述 | The K-ras oncogene is frequently mutated and the p53 tumor suppressor gene is occasionally mutated in various human carcinomas. To determine whether these gene mutations play a role in the development of ovarian carcinomas, we analyzed 175 fresh frozen tissues and 190 formalin-fixed paraffin-embedded tissues. In the present study we examined 175 cases of ovarian tumors for mutations in the K-ras oncogene using the polymerase chain reaction and dot blot hybridization. Mutations in codon 12 of K-ras were detected in 37 of the 175 cases (21.1%). These mutations were found in 10 of the 29 (34.5%) mucinous adenocarcinomas, and 3 of the 4 (75.0%) mucinous cystadenomas (LPM). Immunohistochemical staining for the p53 protein was performed in 200 formalin-fixed paraffin-embedded tissues using monoclonal antibody PAb1801. No staining was seen in benign samples, whereas intense nuclear staining of cancer cells consistent with overexpression of the p53 protein was observed in 29 of the 200 cases (14.5%). In serous adenocarcinoma, p53 overexpression was more frequent in advanced (stage III, IV) cancers (18/36, 50.0%) than in early (stage I, II) cancers (0/20, 0.0%). None of the cases had both point mutation in codon 12 of the k-ras and overexpression of the p53. These findings showed that genetic changes involving protooncogenes or suppressor genes occur during ovarian carcinogenesis. | |||||||||||||
