| 内容記述 | A protein with trypsin inhibitory activity was purified from human urine. The molecular mass weight was about 25,000, and it reacted with anti-κ antibody, but not with anti-λ, anti-γ, anti-α, anti-μ, or anti-urinary trypsin inhibitor (UTI). The N-terminal amino acid sequence of this protein demonstrated on 82% similarity with the corresponding region of human immunogloblin κ chain (REI). Monoclonal κ chain was purified from the urine of a patient with multiple myeloma. The trypsin inhibition of κ chain was competitive with the apparent Ki of 0.376 μM. It inhibited only trypsin and pancreatic kallikrein, but not chymotrypsin, plasmin, plasma kallikrein. neutrophil elastase, pancreatic elastase, thrombin, cathepsin G, thermolysin, collagenase or papain. Urine specimens from 18 patients with mulutiple myeloma were examined, 5 κ chains from 10 specimens had trypsin inhibitory activity, whereas no inhibition was observed in any of 8 λ chains studied. | |||||||||
