We performed comparative studies on conscious restrained 6-month-old female SHRSP and WKY, chronically instrumented for blood pressure (BP) measurement. The potent nitric oxide (NO) inhibitor N^G-nitro-L-arginine (L-NNA, 3 and 6 mg/kg i.v.) produced a dose-related increase in BP with concomitant reduction in heart rate (HR). Systolic arterial pressure (SAP) enhanced more markedly in SHRSP, whereas the decreases in HR were more pronounced in WKY (p<0.05). In the presence of L-NNA, the hypotensive and tachycardic responses to glyceryl trinitrate (GTN, i.v. bolus doses of 3, 10 and 30 μg/kg) were more increased, and the rises in HR were significantly higher in the group of WKY compared to SHRSP. Our findings suggest that L-NNA caused more pronounced effects on the NO-involved system in SHRSP than in WKY. The less bradycardic response in SHRSP compared to WKY after treatment with L-NNA may be caused by disorders in the baroreflex system. The NO-synthase inhibitor, L-NNA, potentiated the vasodilator effect of GTN in WKY as well as in SHRSP rats. |
