内容記述 | Recently, the specific receptor for the urokinase-type plasminogen activator (u-PA) receptor, u-PAR was demonstrated in several cell types. u-PA bound to u-PAR is closely associated with pericellular proteolysis. In the present study, the effect of bone resorbing factors on the pericellular fibrinolytic system of osteosarcoma NY cells was investigated. NY cells secrete single-chain u-PA (scu-PA) which is converted to two-chain u-PA by several proteases such as plasmin Parathyroid hormone (PTH), prostaglandine E_2 (PGE_2) or tumor necrosis factor α (TNF-α) enhanced the secretion of u-PA antigen and u-PA activity, and suppressed the secretion of plasminogen activator inhibitor-1 (PAI-1) antigen to the conditioned medium. The former two factors also increased u-PA antigen and u-PA activity on the cell surface. Transforming growth factor β (TGF-β) enhanced the u-PA antigen in the conditioned medium, but its activity was suppressed due to the increased secretion of PAI-1. ^<125>I-DFP-u-PA bound specifically to NY cells and the binding analysis revealed the presence of a single class of binding sites for u-PA. PTH and PGE_2 increased Bmax by 1.4-fold and 1.7-fold, respectively, and enhanced the u-PAR mRNA levels by 1.4-fold and 2.4-fold, respectively. However, TGF-β did not alter either the Kd or u-PAR mRNA level. Thus, pericellular fibrinolytic activity by u-PA/u-PAR and PAI-1 is modulated by bone resorbing factors. | |||||||||
