Keratan sulfate (KS) production was measured in parallel with PGE2 release in chondrocytes. IL-1 inhibited KS production and stimulated PGE2 release. Indomethacin and dexamethasone partially blocked the IL-1-induced PGE2 release and allowed the IL-1 inhibited KS production recover. In the presence of PGE2, there was a dose-dependent decrease in baseline KS production. The protein kinase C inhibitor H7 did not block the actions of IL-1, whereas the cyclic nucleotide-dependent protein kinase inhibitor H8 blocked PGE2 enhancement and KS production.
