内容記述 Contractions due to release of intracellular stored Ca in smooth muscle of resistance vessels from stroke-prone spontaneously hypertensive rats (SHRSP) and normotensive control Wistar Kyoto rats (WKY) were studied. Caffeine- and noradrenaline-induced contractions were gradually decreased by the removal of extracellular Ca, and no contraction could be induced 60 min after the removal of Ca from preparations of both SHRSP and WKY. High-K-induced contractions were abolished within 10 min indicating that the extracellular Ca was removed within this period. Both caffeine- and noradrenaline-induced contractions observed 10 min after the removal of extracellular Ca were greater in preparations from SHRSP. The amount of Ca released by caffeine and noradrenaline in the presence of Ca was estimated by extrapolating the time course of decay back to time 0. It was shown that the estimated Ca-release in the presence of 2 mM Ca was greater in the preparation from SHRSP than that from WKY. Caffeine-induced contraction could be blocked by procaine or ryanodine. An observation using fura 2, a Ca indicator, revealed that caffeine and noradrenaline induced a greater increase in intracellular Ca in the preparation from SHRSP. The extracellular Ca-independent component of the phorbol ester (TPA)-induced contraction was identical, indicating no difference in the contractile apparatus in smooth muscle. These results suggest that the greater contractile response of caffeine and noradrenaline were due to greater release of Ca from sarcoplasmic reticulum, i. e. both Ca- and IP_3- induced Ca-release are increased in smooth muscle of the mesenteric artery of SHRSP compared with that of WKY.
