内容記述 Insulin-dependent diabetes mellitus (IDDM) is an autoimmune disorder associated with a selective destruction of pancreatic beta cells. Non-obese diabetic (NOD) mice which spontaneously develop diabetes have been established as an animal model of IDDM. The present study investigated the in vitro production of interleukin 6 (IL-6), a potent differentiation factor of B-lymphocytes, by spleen cells in NOD mice in order to examine how IL-6 is related to the development of diabetes in NOD mice. IL-6 was measured by the bioassay using an IL-6 dependent murine hybridoma cell line (7TD1). The maximum response of the in vitro IL-6 production by spleen cells was obtained after the incubation of spleen cells with 25μg/ml of lipopolysaccharide (LPS) for 72 h. In the study of age effect of LPS-induced IL-6 production by spleen cells, NOD mice showed two peaks at 4 and 16-18 weeks of age, while control ICR mice showed simply an age-related decrease for the production. Since NOD mice develop spontaneous diabetes after 20 weeks of age, the precedent increase of IL-6 production specific to NOD mice suggests a pathophysiological significance of IL-6 in the diabetes of NOD mice. In accordance with this, the IL-6 production in NOD mice with diabetes was higher than that in NOD mice without diabetes. Also, NOD mice with newly developed diabetes showed higher IL-6 production than those with long term diabetes. Cyclophosphamide (CY) which, is known to accelerate the development of diabetes in NOD mice was given to NOD and ICR mice at 15 weeks of age. One and two weeks after the treatment, the LPS-induced IL-6 production in spleen cells was significantly increased in NOD mice but not in ICR mice. In conclusion, IL-6 production seems to be closely associated with the development of diabetes in NOD mice.
