内容記述 | The effects of Isoteoline (IST), a new tetrahydroaporphine derivative, on the blood pressure and renin-angiotensin-aldosterone system (RAAS) in spontaneously hypertensive rats (SHR) were studied. The experiments were performed with adult (8 months of age) male SHR of the Okamato-Aoki strain, generation F^<26>-F^<37> weighing 250±10g. Systolic blood pressure was measured indirectly in conscious restrained rats by tail-cuff plethysmography with sphygmomanometer. Plasma renin activity, angiotensin II and aldosterone plasma levels were determined by radio-immunoassay. The angiotensin converting enzyme (ACE) activity in vivo and in vitro was estimated by a modified fluorometric assay, while changes in the electrolyte levels in plasma (Na^+, K^+ and Cl^-) were determined by flame photometry. It was established that IST administered in doses of 1 and 5 mg/kg orally and intraperitoneally after single (6 days) and long-term (14 days 3 times daily) treatment decreased the systolic blood pressure and levels of the components of RAAS. The results suggest that the effects of IST on SHR was most probably realized through an influence on the adrenergic and dopaminergic mediatory systems. | |||||||||||||||
